  • """State transitions"" u cyanobakterií je mechanismem fyziologické adaptace, která změní interakci fykobilisomu s komplexem fotosystémemů I a II. Tento mechanismus je nezbytný pro cyanobakterie při nízké intenzitě světla. Studie náhodných mutací u cyanobakterií identifikovaly gen rpaC, který je specificky potřebný pro ""state transitions"". Dřívější studie ukázaly, že gen rpaC je esenciální pro životaschopnost buňky u kmene Synechocystis sp. PCC6803 a produkt genu rpaC je s vysokou pravděpodobností strukturní složkou fykobilisom-fotosystém II superkomplexu. Avšak fyziologická role proteinu je nejasná. Cílem tohoto projektu je purifikace rekombinantního proteinu kodovaného rpaC genem a určení struktury a konformace pomocí biofyzikálních metod. Výsledné strukturní informace pomohou při objasnění mechanismu ""state transitions""." (cs)
  • State transitions in cyanobacteria are physiological adaptation mechanism that change the interaction of the phycobilisomes with the Photosystem I a Photosystem II core complexes, and this mechanism is essential for cyanobacterial at very low light intensities. A random mutagenesis study in cyanobacterium identified a gene named rpaC, which appeared to be specifically required for state transitions. A previous study shown that rpaC gene is essential for cell viability in Synechocystis sp. PCC6803, and gene product of rpaC is with high probability structural component of phycobilisome-Photosystem II supercomplex. However, the physiology role of the protein is unclear. The aim of this project is purified recombinant protein encoded by rpaC gene and determined the protein structure and conformation by biophysical methods. Resulting structural information will help to elucidate mechanism of state transitions. (en)
