  • Poslední krok v biosyntéze c cytochromů, které jsou nepostradatelné jak pro fotosyntézu tak pro respiraci, zahrnuje transport hemu přes membránu a jeho kovalentní připojení k apoproteinu. Tři rozdílné biosyntetické dráhy se vyvinuly u různých organismů. Nejméně je známo právě o tomto procesu u sinic a chloroplastů. Naše výsledky získané u sinice Synechocystis sp. PCC 6803 ukazují, že dva proteiny CcsA a CcsB mohou hrát centrální roli v tomto procesu, kde CcsA bude vázat hem a CcsB apoprotein. Cílem tohoto ptojektu je otestovat tento model maturace c cytochromů u sinic, zvláště pak roli CcsB. Zkoumány budou dvě hlavní otázky: 1) Je CcsB chaperonem apocytochromu 2(Tvoří CcsB komplex s CcsA a tvoří tento komplex jádro maturačního komplexu. Použitá metodologie bude zahrnovat overexpresi obou proteinů, izolaci polyklonálních protilátek a detekci obou komplexů in vivo. (cs)
  • The last step for biosynthesis of c type cytochromes, indispensable for both photosynthesis and respiration, involves heme transport across the membrane and its covalent attachment to the apoprotein. Three differenc pathways evolved among various organisms. The pathway used in cyanobacteria and chloroplasts is the least studied. Our results from cyanobacterium Synechocystis sp. PCC 6803 indicate that two proteins CcsA and CcsB may play a central role in this process with CcsA binding heme and CcsB binding apocytochrome. This research project is aimed to test this model of cytochrome c maturation in cyanobacteria, particularly the role of the CcsB protein in it. Two major problems will be investigated: 1) Does CcsB serve as apocytochrome chaperone 2) Does CcsB form a complex with CcsA, does this complex form the core of the maturation complex. Tethodology is based on overexpression of both proteins, generation of polyclonal antibodies and detection of both complexes in vivo. (en)
