Description
| - Jaderné receptory tvoří rodinu transkripčních faktorů zapojených do regulace nejrůznějších biologických procesů. K identifikaci genů, jež jsou regulovány tyroidálním hormonem, jsme použili systém diferenciace prekurzorů červených krvinek akcelerováné tyroidálním hormonem. Jedním z genů, které jsou přímo regulovány tyroidálním hormonem v tomto systému je TRUP1, nový člen rodiny bílkovin obsahujících SSD doménu. Klonováním jsme získali plnodélkovou cDNA kódující bílkovinu o velikosti 150 kDa, která obsahuje 13 transmembránových domén. TRUP1 je exprimován v mozku, srdci, sítnici a varlatech. Cílem našeho projektu je přesně popsat subcelulární lokalizaci bílkoviny, definovat její expresi v různých tkáních a buněčných typech a potvrdit její regulaci tyroidálním hormonem in vivo. Dále budeme studovat možnou interakci mezi bílkovinou TRUP1 a cholesterolem, čímž chceme přispět k objasnění vztahu mezi tyroidálním hormonem a metabolismem cholesterolu na molekulární úrovni. (cs)
- Nuclear receptors comprise a family of transcription factors that are involved in the regulation of diverse biological processes. We have used the model of thyroid hormone (T3)-accelerated erythroid differentiation to search for thyroid hormone receptor-regulated genes. We have identified TRUP1, a new member of the sterol-sensing domain protein family, as a gene directly regulated by ligand-activated thyroid hormone receptor. Cloning of the full-length cDNA revealed that it encodes a protein of 150 kDa containing 13 transmembrane domains. It is expressed in brain, heart, retina and testis. During the proposed project we wish to precisely describe the subcellular localization of TRUP1, define its expression pattern in different tissues and cell types and confirm its T3 regulation in vivo. We will attempt to study potential physical interaction between TRUP1 and cholesterol and also to identify the molecular link between T3 action and cholesterol metabolism at the cellular level. (en)
