Procathepsin D (pCD) plays a critical role in breast cancer: it exerts mitogenic activity toward breast cancer cells, promotes tumor progression and metastasis. The pCD mitogenicity is mediated via interaction with a proliferative receptor on breast cancer cells. The research aims are focused on discovery of chemical tools for the regulation of pCD binding to this receptor. First, the receptor will be identified through proteomic analysis of protein-protein interactions. Second, the receptor-binding domain of pCD will be localized by peptide mapping and mutation analysis. Small mimetics of this domain will be designed using combinatorial chemistry and molecular modeling to derive the antagonist compound with antiproliferative activity. Third, sphingolipids and glycosaminoglycans will be screened for modulation of pCD function. The project will bring new molecules regulating the pCD mitogenicity as leads for the development of potential anticancer chemotherapeutics. (en)
Prokatepsin D (pCD) hraje zásadní úlohu při nádorovém onemocnění prsu: má mitogenní účinek na nádorové buňky, podporuje růst nádoru a metastázu. Mitogenní aktivita je zprostředkována jeho interakcí s proliferačním receptorem na nádorových buňkách. Projekt je zaměřen na vývoj nových chemických nástrojů pro regulaci vazby pCD na tento receptor. (1) Receptor bude identifikován proteomickou analýzou protein-proteinových interakcí. (2) V molekule pCD bude pomocí peptidového mapování a mutační analýzy lokalizována vazebná doména pro receptor. S využitím kombinatorní chemie a molekulárního modelování budou navržena malá antagonistická mimetika s antiproliferační aktivitou. (3) Sfingolipidy a glykosaminoglykany budou testovány jako potenciální modulátory pCD. Projekt přinese nové molekuly regulující mitogenní aktivitu pCD a umožňující vývoj nových protinádorových chemoterapeutik. (cs)
