Description
  • Sperm maturation in the epididymis represents a key step in the reproduction process. Sperm plasma membrane is exposed to epididymal fluid representing the natural environment essential for their post-testicular maturation. Changes in the sperm membrane proteins are influenced by proteolytic and glycosidic enzymes present in the fluid. We monitored acrosin inhibitor (AI) in epididymal fluid and spermatozoa along the organ. Boar epididymal fluid with spermatozoa was isolated according to its spatial restriction into 9 fractions. The AI was detected in sperm extracts and fluids using specific polyclonal antibody. Expression of AI gene was investigated by PCR method. Additionally, immunofluorescence technique (IMF) was used for localization in the tissue. We registered the increasing signal of AI from caput to cauda epididymis. We confirmed that using gene expression method. Using specific antibody, AI was localized in the secretory tissues of caput, corpus and cauda epididymis by IMF.
