  • The main role of proteinase inhibitors in the reproductive tract is to inactivate prematurely released hydrolytic enzymes from damaged spermatozoa and protect the male and female reproductive tracts and spermatozoa against proteolytic degradation. We studied the expression and localization of acrosin inhibitor isolated from seminal plasma and spermatozoa in the boar reproductive tract. The amino acid sequence of boar acrosin inhibitor and its biochemical properties were completely determined. However, its origin and localization have not been fully elucidated. Acrosin inhibitor mRNA was detectable in epididymis, seminal vesicles, prostate and Cowper’s glands. Polyclonal antibody against purified acrosin inhibitor detected and localized this protein in extracts and secretory tissues of boar reproductive organs. The acrosin inhibitor was immunodetected in epididymis, prostate and seminal vesicles, and additionally in sperm extracts and on epididymal and ejaculated spermatozoa.

  • Hlavní rolí proteinasových inhibitorů v reprodukčním traktu je inaktivovat předčasně uvolněné hydrolytické enzymy z poškozených spermií a chránit tak samčí i samičí reprodukční trakt před proteolytickou degradací. Studovali jsme expresi a lokalizaci inhibitoru akrosinu izolovaného ze semenné plasmy a spermií. Aminokyselinová sekvence a biochemické vlastnosti tohoto inhibitoru byly určeny, ale původ a lokalizace v reprodukčním traktu nebyly zcela vyřešeny. mRNA tohoto inhibitoru byla detekovana v epididymu, semených vacích, prostatě a Cowperových žlázách. Polyklonální protilátka proti přečištěnému akrosinovému inhibitoru detekovala a lokalizovala tento protein v extraktu a sekrečních tkáních kančích reprodukčních orgánů. Inhibitor akrosinu byl detekován protilátkou v epididymu, prostatě a semenných vacích, ale také v extraktech ze spermií a na povrchu kančích epididymálních a ejakulovaných spermií. (cs)
