Description
  • Differentiation of hematopoietic stem cells and progenitors into various lineages is controlled by a complex array of extrinsic and intrinsic factors. Here we have identified, cloned and characterized chicken thrombopoietin, the first non-mammalian Tpo and its receptor c-Mpl. Chicken Tpo is expressed in the liver and spleen whilst c-mpl is expressed in the bone marrow, lung and spleen. In vitro experiments with sorted multipotent bone marrow-derived progenitors demonstrate the committing role of Tpo for the thrombocytic lineage. In cooperation with stem cell factor Tpo also plays a role in self-renewal of multipotent progenitors. Intravenous application of recombinant chicken Tpo or overexpresion of Tpo and c-mpl via retroviral infection lead to erythroblastosis and thromboblastosis in animals. In this work we have introduced the experimental model of the chicken bi-potent thrombo-/erythropoietic progenitor that can be useful for identifying key regulators of cell fate determination.

  • Diferenciace hematopoetických kmenových buněk a progenitorů do různých linií je kontrolována řadou vnějších a vnitřních faktorů. Identifikovali, klonovali a charakterizovali jsme kuřecí trombopoetin, první nesavčí Tpo a jeho receptor c-Mpl. Kuřecí Tpo se exprimuje v játrech a slezině, zatímco c-mpl se exprimuje v kostní dřeni, plicích a slezině. Pokusy in vitro ukazují, že Tpo směruje multipotentními progenitory z kostní dřeně do trombocytární linie. Tpo společně s SCF hraje svou roli v sebeonově multipotemtních progenitorů. Nitrožilní aplikace rekombinantního kuřecího Tpo nebo nadexprese Tpo a c-mpl pomocí retrovirů vede k erytroblastose a tromboblastose u zvířat. V této práci jsme zavedli model kuřecích bipotentních trombo/erytropoetických progenitorů které mohou být užitečné pro identifikaci klíčových regulátorů buněčného osudu. (cs)
