  • Colorectal tumors arise from unregulated cell proliferation of the intestinal epithelium through a multistep process, with premalignant adenomas as first step. Familial adenomatous polyposis patients carry a germline mutation in the APC gene leading to development of thousands of polyps, which when untreated lead to cancer. This study aimed to establish conditions for culture of epithelial cells composing an adenomatic structure. All colorectal specimens were obtained from FAP patients within 1-2 hrs of surgery. Cells were cultured by standard proceduresand characterized by immunostaining with pancytokeratin, vimentin and desmoplakin antibodies. Successful subculturing of the cell sheets took place only when dispase prepared in Ca2+ and Mg2+ free medium, was used to digest polyp tissue taken from FAP patients. By using immunostaining these cells were characterized as epithelial. The protocol we developed here provides a means of preparing cell cultures from human colorectal adenomas.

  • Kolorektální nádory vznikají z neregulované buněčné proliferace střevního epitelu vícekrokovým procesem. jehož prvním krokem je obvykle vytváření premaligních adenomů. Pacienti s familiární adenomatózní polypózou mají dědičné mutace v genu APC vedoucí ke vzniku tisíce polypů, které, pokud zůstanou bez ošetření, mohou vést k rakovině. Cílem této studie bylo vytvoření podmínek pro kultivaci epiteliálních buněk z adenomů FAP pacientů. Všechny explantáty adenomů byly získány z FAP pacientů 1-2 hod po operaci. Buňky byly kultivovány standardním procedurami a charakterizovány imunochemicky pomocí pancytokeratin, vimentin a desmoplakin protilátek. Úspěšné subkultury epiteliálních buněk adenomu bylo dosaženo pouze v případě, kdy pro rozvolnění polypové tkáně FAP pacientů byla použita dispáza připravená v Ca2 + a Mg2 + neobsahujícím médiu. Imunochemicky byly tyto buňky charakterizovány jako epiteliální. Vyvinutý protokol umožňuje přípravu buněčných kultur z lidských kolorektálních adenomů. (cs)
