  • Taurine is a widespread natural product whose nitrogen moiety was recently shown to be assimilated by bacteria, usually with excretion of an organosulfonate via undefined novel pathways. A screen of genome sequences allowed the hypothesis that the marine gammaproteobacterium Neptuniibacter caesariensis would inducibly assimilate taurine-nitrogen and excrete sulfoacetate. The pathway involved an ABC transporter, taurine:pyruvate aminotransferase (Tpa), a novel sulfoacetaldehyde dehydrogenase (SafD) and exporter(s) of sulfoacetate. Inducible activities of Tpa and SafD were detected. SafD was purified to homogeneity. SafD activity was found specific for sulfoacetaldehyde. The N-terminal amino acid sequence of SafD confirmed the identity of the safD gene. The pathway genes were transcribed inducibly, which indicated expression of the whole hypothetical pathway. We presume that this pathway is one source of sulfoacetate in nature, where this compound is dissimilated by many bacteria.

  • Taurin je rozšířený přírodní produkt, jehož aminoskupina je asimilována bakteriemi (obvykle za exkrece organosulfonátu) prostřednictvím nových, dosud nedefinovaných metabolických drah. Na základě analýzy genomových sekvencí byla vytvořena hypotéza, že mořská gammaproteobakterie Neptuniibacter caesariensis je schopná induktivně asimilovat taurin jako zdroj dusíku za současné exkrece sulfoacetátu. Metabolická dráha zahrnovala ABC transportér, taurin:pyruvát aminotransferasu (Tpa), nový enzym sulfoacetaldehyd dehydrogenasu (SafD) a exportér(y) vznikajícího sulfoacetátu. Indukovatelná aktivita Tpa a SafD byla detekována. SafD byla purikována do homogenity. Aktivita SafD byla specifická pro sulfoacetaldehyd. Sled aminokyselin na N-konci SafD se shodoval se sekvencí genu safD. Geny dráhy byly transkribovány induktivně, což svědčí o expresi celé metabolické dráhy. Předpokládáme, že tato dráha je jeden ze zdrojů sulfoacetátu v přírodě, kde je tato sloučenina disimilována mnoha bakteriemi. (cs)
