  • Adult bone mineral density (BMD), an important risk factor for fracture, is the result of genetic and environmental interactions. A Quantitative Trait Locus (QTL) for the phenotype of volumetric (v)BMD, named Bmd8, was found on mid-distal Chromosome (Chr) 6 in mice. This region is homologous to human Chr 3p25. The B6.C3H-6T (6T) congenic mouse was previously created to study this QTL. Utilizing block haplotyping of the 6T congenic region, expression analysis in the mouse, and by examination of non-synonymous Single Nucleotide Polymorphisms (SNP), Peroxisome Proliferator Activated Receptor Gamma (Pparg) was determined to be the most likely candidate gene for the Bmd8 QTL out of the 630 genes located in the congenic region. The findings suggest that dietary fat has a significant influence on BMD that is dependent on the alleles present for the PPARG gene.

  • Kostní densita dospělých jedinců (BDM) jako důležitý rizikový faktor pro zlomeniny je důsledkem genetických a enviromentálních interakcí. QTL lokus pro volumetrický BDM fenotyp, Bmd8, byl nalezen na distálním chromosomu (Chr) 6 u myší. Tato oblast je homologní lidskému Chr 3p25. B6.C3H-6T (6t) kongení myš byla vytvořena minule ke studiu tohoto QTL. Využitím mapování haplotypových bloků v 6T oblasti, extersního profilování u myší a vyštřením nesynonymních SNPs, Peroxisome Proliferator Activated Receptor Gamma (Pparg) byl stanoven jako nejpravděpodobnější kandidát genu pro Bmd8 QTL ze 630 genů umístěných v kongenní oblasti. Toto zjištění naznačuje, že dietetický tuk má signifikantní vliv na BMD, který je závsilý na alelách přítomných pro PPARG gen. (cs)
