  • Signální dráha proteinu Wnt ovlivňuje proliferaci a udržování hemapoetických buněk, kdežto cytotoxický ligand Apo2L/TRAIL navozuje apoptózu různých transformovaných buněk. Kokultivací lidských pre-B leukemických buněčných linií KM3 nebo REH spolu s krysími embryonálními fibroblasty, jež produkují proteiny Wnt1 nebo Wnt3a, byla úspěšně potlačena Apo2L/TRAIL způsobená apoptóza leukemických buněk. Tato suprese se děje na úrovni ranných stadií Apo2L/TRAIL indukované apoptotické kaskády, přičemž samotné spuštění signalizace proteinu Wnt v lidských leukemických buňkách nevede k dostatečné ochraně proti apoptóze. Inhibice MEK1/MEK2 nebo NFkB signální dráhy v pre-B liniích KM3 a REH zrušila antiapoptotický efekt Wnt produkujích fibroblastů. Tyto výsledky naznačují, že parakrinní signály spojené s dráhou proteinu Wnt mohou být důležité pro přežívání pre-B leukemických buněk. (cs)
  • The Wnt signaling enhances proliferation and maintenance of hematopoietic cells. In contrary, the cytotoxic ligand Apo2L/TRAIL induces apoptosis of transformed cells. We observed that pre-cultivation of pre-B leukemia cell lines (KM3, REH) on Wnt1- or Wnt3a-expressing Rat2 fibroblasts efficiently suppressed their Apo2L/TRAIL -induced apoptosis. This suppression occurs at the early stages of the Apo2L/TRAIL apoptotic signaling and is only partially dependent on the Wnt pathway alone. Anti-apoptotic Rat2Wnt1 signaling was significantly hampered by the inhibition of MEK1/MEK2 or NFkB pathways in pre-B acceptor cells. Our results implicate the existence of an additional signaling pathway in Wnt1-transfected fibroblasts that in conjunction with the Wnt-triggered cascade of cellular events suppresses Apo2L/TRAIL-induced apoptosis of pre-B cells. Such composed antiapoptotic signaling could contribute to pre-B leukemias survival.
