Description
  • Neurogenesis in developing neocortex is strictly regulated. We report gradual retreat of canonical Wnt signaling in the cortex from lateral-to-medial and anterior-to-posterior as a neurogenesis prerequisite. Ectopic expression of a beta-catenin/LEF1 fusion protein maintains active canonical Wnt signaling in developing cortex and delays expression onset of neurogenic factors Pax6, Ngn2, Tbr2, and subsequent neurogenesis. Contrary to this, conditional ablation of beta-catenin accelerates expression of the same neurogenic genes. We also show sustained canonical Wnt activity in the lateral cortex to give rise to cells with hippocampal characteristics in the cortical plate at the expense of cortical fate and to cells with dentate gyrus characteristics in hippocampus. The canonical Wnt signaling dose seems to determine cellular fate in developing cortex and hippocampus and recession of Wnt signaling seems to act as a morphogenetic gradient regulating neurogenesis in the cortex.

  • Neurogeneze je přísně regulovaný proces buněčného dělení a diferenciace. Zde jsme prokázali, že postupný úbytek kanonické wnt signalizace v mozkové kůře je předpokladem pro neurogenezi. Navíc, naše výsledky ukazují, že trvalé zapnutí canonical wnt aktivity v kůře dává vzniknout buňkám s hippocampálními vlastnostmi. To naznačuje, že dávka wnt signálu určuje buněčných osud v vyvíjející se kůře a hippokampu. (cs)
