  • Mature spermatozoon exhibits extraordinary structural compartmentalization due to the presence of cytoskeletal proteins and functions in fertilization and motility. We have shown previously that anti-β-tubulin antibody TU-12 provided unexpectedly strong reaction in human and boar sperm head. Now we wanted to determine location of the corresponding epitope on the tubulin molecule and to find any differences in its exposition in human sperm with normal and pathological spermiograms. Sperm were divided into 5 categories: normozoo-, oligozoo-, asthenozoo-, teratozoo- and astheno-teratozoospermia, and well-characterized monoclonal antibodies were applied to monitor tubulin epitope distributions. TU-12 epitope was located in β-tubulin region β426-435. Immunoblotting revealed differences in the amount of tubulin among men with normal and pathological spermiograms. Striking differences were observed especially in sperm heads. Tubulin epitopes could thus be useful biomarkers of sperm pathology.

  • Zralé spermie mají jasnou strukturu, která je ve vztahu k cytoskeletálním proteinům, které mají funkční úlohu v procesu fertilizace a motilitě spermií. V předchozích publikacích jsme prokázali neočekávaně jasnou detekci β-tubulin v hlavičce kančích a lidských spermií pomocí protilátky TU-12. V této práci byl jasně detekován epitop tubulinové molekuly a rozdíly v jeho detekci u lidských spermií s normálním a patologickým spermiogramem. Spermie byly rozděleny do pěti kategorií normozoospermia, oligozoospermia, asthenozoospermia, teratozoospermia a astheno-teratozoospermia. TU-12 epitop byl lokalizován v β-tubulinové oblasti β426-435. Immunobloting prokázal kvantitativní rozdíly v množství tubulinu mezi muži s normálním a patologickýcm spermiogramem. Podobně byly prokázany i rozdíly v expozici TU-12 epitopu v hlavičce spermií u normálních a patologických spermií. Výsledky prokázaly možnost použití tubulinového epitopu jako biomarkeru pro stanovení patologického stavu spermií. (cs)
