  • c-Myb, known to play a central role in hematopoiesis, is also an important factor involved in myogenesis. Here, we found that both c-myb and its alternatively spliced form c-mybE9A are expressed in proliferating C2C12 myoblasts and turned off in differentiating cells. Constitutive expression of exogenous c-myb in C2C12 cells inhibits their terminal differentiation. It is shown that the c-Myb protein physically interacts with MyoD, the key regulator of myogenesis, and inhibits MyoD-dependent transcription. The interaction domains are the DNA binding domain of c-Myb and the bHLH motif of MyoD. Our data suggest that in proliferating cells c-Myb binds MyoD and inhibits its transcriptional activity until cell-cell contacts are established and c-myb expression is switched off. Thus, the c-Myb protein may be one of factors ensuring that proliferating myoblasts remain undifferentiated.

  • c-Myb, o němž je známo že hraje centrální roli v krvetvorbě, je také důležitý faktor v myogenezi. Zjistili jsme, že jak c-myb, tak jeho alternativně sestřižená forma c-mybE9A je exprimována v dělících se C2C12 myoblastech, v diferencujících buňkách je jejich exprese vypnuta. Vynucená exprese vloženého c-myb genu zablokuje diferenciaci C2C12 buněk. Je ukázáno, že c-Myb protein se fyzicky váže na MyoD, hlavní faktor regulující myogenezi a inhibuje transkripci, která je závislá na MyoD. Vazebné oblasti jsou DNA vazebná doména c-Mybu a bHLH oblast MyoD. Na základě našich výsledků předpokládáme, že c-Myb se váže na MyoD a inhibuje jeho aktivitu až do doby než se buňky dostanou do vzájemného kontaktu a exprese c-mybu se vypne. c-Myb tak může být jedním z faktorů zajištující že dělící se myoblasty nemohou diferencovat.
