  • Neural progenitors of mouse forebrain can be propagated in vitro with bFGF and EGF as neurospheres. Less is clear if regional or developmental stage properties of these cells are kept in neurosphere cultures. We show that their original cell fate is lost. We isolated neural progenitors from dorsal telencephalon of D6-GFP mice for in vitro culture. The expression profile was specifically changed in cultured cells in 3 passages. Markers of the dorsal forebrain were downregulated and several ventrally-expressed genes were induced. Altered gene expression led to deep phenotypic change of cultured cells. D6-GFP+ cortical progenitors produce excitatory neurons in the cortex and few astrocytes in vivo but after culture in vitro they differentiate into many astrocytes, and oligodendrocytes and inhibitory neurons. Wnt signaling in cultured neurospheres was downregulated similarly as other dorsal markers but dominant active Wnt signaling retarded the loss of the dorsal identity in neurospheres.

  • Nervové kmenové buňky (NSC) jsou univerzální buňky, které lze kultivovat in vitro v chemicky definovaném médiu s růstovými faktory FGF a EGF. Je možné navodit diferenciaci těchto buněk na základní buněčné typy přítomné v centrálním nervovém systému: neurony, astrocyty a oligodendrocyty. NSC je možné získat jak z embryonální nervové tkáně, tak z mozku dospělců. Otázkou zůstává, zda si izolované NSC uchovávají vlastnosti buněk dané oblasti mozku, z které byly získány. V této práci jsme použili transgenní myší linii D6-GFP, v které jsou neurony šedé kůry mozkové označeny fluorescenčním proteinem GFP. Izolovali jsme embryonální NSC pomocí fluorescenční metody FACS a buňky kultivovali in vitro. Pomocí buněčných markerů, real-time PCR a Affymetrix analýzy jsme ukázali zásadní změny vlastností diferencovaných buněk oproti jejich původnímu fenotypu. (cs)
