  • Produkce rekombinantního lidského erytropoietinu (rhEPO) pro léčebné účely je založena ne jaho expresi ve vybraných klonech transfekovaných savčích buněk. Tento glykoprotein může být také produkován cílenou sekrecí do tělesných tekutin transgenních zvířat. Zde popisujeme přípravu transgenních králíků produkujících rhEPO do laktující mléčné žlázy. Transgenní jedinci jsou životaschopní, plodní a předávají gen rhEPO potomstvu. Údaje z Northernova přenosu ukázaly, že exprese transgenu v mléčné žláze je během laktace řízena regulačními sekvencemi WAP. Zatímco hybridizace s celkovou RNA ukázala expresi pouze v laktující mléčné žláze, vysoce citlivý kombinovaný přístup využívající techniku RT-PCR/hybridizace odhalil slabou ektopickou expresi. Úroveň sekrece rhEPO v zakladatelské transgenní samici měřená v době laktace se pohybovala v rozmezí 60-178 a 60-162 mUI/ml v mléku resp. krevní plazmě. Biologická aktivita rhEPO z mléka byla potvrzena standardním testem inkorporace 3H-thymidinu. Popi... (cs)
  • Production of recombinant human erythropoietin (rhEPO) for therapeutic purposes relies on its expression in selected clones of transfected mammalian cells. Alternatively, this glycoprotein can be produced by targeted secretion into the body fluid of transgenic mammals. Here we report on the generation of a transgenic rabbits producing rhEPO in the lactating mammary gland. Transgenic individuals are viable, fertile and transmit the rhEPO gene to the offspring. Northern blot data indicated that the expression of the transgene in the mammary gland is controlled by WAP regulatory sequences during the period of lactation. While the hybridization with total RNA revealed the expression only in the lactating mammary gland, the highly sensitive combinatory approach using RT-PCR/hybridization technique detected a minor ectopic expression.
