  • Whey acidic protein, WAP, is being recognized as a principal milk protein expressed in pregnant or lactating females of the several mammalian species. Recently it has been shown that 6.3kb 5Ž-untranslated region of the rabbit WAP (rWAP) gene is able to control, and almost completely restrict, the expression of transgene into the mammary gland of the transgenic animal. We cloned genomic fragment carrying the rWAP gene locus from the ?DASHII phage genomic library and used 8.5 kb long 5Ž-untranslated part of the rWAP gene to target the expression of the human erythropoietin gene (hEPO), cloned from the ?FIXII phage genomic library, into the mammary gland of the mouse. Vectors, carrying the hEPO gene or the rWAP-hEPO hybrid gene construct, were injected into the mice ova and 12 transgenic animals were identified by PCR and Southern blot from the progeny of 168 tested littermates.   • Kyselý protein syrovátky, WAP, se považuje za nejdůležitější mléčný protein exprimovaný v březích či kojících samic u řady savčích druhů. Nedávno bylo prokázáno, že 5´-netranslatovaná oblast králičího genu pro WAP (rWAP) o 6,3 kb může řídit téměř zcela omezit expresi transgenu v mléčné žláze trangenního zvířete. Klonovali jsme fragment genomu obsahující lokus genu rWAP z fágové genomové knihovny ?DASHII a využili jsme 8,5 kb dlouhou 5´-netranslatovanou část genu rWAP k cílení exprese lidského genu pro erytropoietin (hEPO), klonovaného z fágové genomové knihovny ?FIXII, do mléčné žlázy myši. Vektory obsahující gen pro hEPO či hybridní genový konstrukt rWAP-hEPO byly injikovány do myších vajíček a pomocí PCR a Southernova přenosu bylo mezi 168 testovanými potomky identifikováno 12 transgenních zvířat. Transgenní myší byly životaschopné, plodné a normálně se vyvíjely. Rekombinantní lidský erytropoietin byl produkován v mléce transgenní myší samice na úrovni sekrece 5,345 mIU/ml, jak by... (cs)
