  • Nově připravené trojrozměrné polymerní nosiče připravené polymerací 2-hydroxyetylmetakrylátu nebo 1-vinyl-2-pyrrolidonu byly modifikovány přítomností kopolymerů, síťovadel a porogenů v různém hmotnostním obsahu, lišily se pak v povrchovém náboji a jeho míře, elastičnosti nebo ve velikosti a četnosti pórů. Následně byly na povrchu i uvnitř polymerních hydrogelů kultivovány myší embryonální kmenové buňky linie D3. Využitím metod RT-PCR, Western blotu, imunohistochemické detekce traskripčního faktoru Oct-4, měření proliferační aktivity a dalších byly nově připravené nosiče testovány z hlediska biokompatibility a použitelnosti pro trojrozměrnou in vitro kultivaci buněk, dále byl pozorován vliv různých modifikací polymeru na nediferencovaný charakter embryonálních kmenových buněk a výskyt buněk uvnitř polymerních hydrogelů.   • Newly synthesized polymer scaffolds based on 2-hydroxyethyl methacrylate (HEMA) and 1-vinyl-2-pyrrolidone (VP) were modified by presence of different weight volumes of copolymeres, crosslinking agents and porogens. Hydrogels differed in surface charge, elasticity or size and count of pores. Subsequently, mouse embryonic stem cells - D3 line (ES D3) - were cultivated on the surface and inside of these hydrogels. Using methods RT-PCR, Western blot, imunohistochemical detection of transcription factor Oct-4, measurement of proliferating activity and others the new prepared hydrogels were tested for their biocompatibility and applicability for the three-dimensional cell growth. Next, it was observed influence of polymer modifications on undifferentiated growth of ES D3 cells and appearance of cells inside of polymer hydrogels.
