  • We studied whether superparamagnetic nanoparticles (CD34 MicroBeads), used clinically for specific magnetic sorting, can be used as a magnetic cell label for in vivo cell visualization. Purified CD34+ cells were implanted into rats with a cortical photochemical lesion, contralaterally to the lesion. On T2W images we observed a hypointensive signal at the injection and lesion site. Prussian blue and anti-human nuclei staining confirmed the presence of magnetically labeled human cells in the corpus callosum and in the lesion four weeks after grafting. Human was detected in brain tissue sections from the lesion using PCR. Our results show that CD34 MicroBeads superparamagnetic nanoparticles can be used as a magnetic cell label for in vivo cell visualization.

  • Sledovali jsme osud CD34 pozitivních lidských progenitorových buněk (h CD34+) značených během imunomagnetické selekce magneticky značenou protilátkou transplantovaných potkanům s korovou fotochemickou lézí. EKB byly injikovány intracerebrálně, do kontralaterální hemisféry. Buňky byly sledovány magnetickou resonancí. Elektronový mikroskop potvrdil přítomnost nanočástic železa na povrchu buněk. Implantované buňky se na NMR zobrazovaly jako hypointenzivní oblasti v okolí vpichu a léze. Přítomnost CD34+ byla potvrzena histologicky, barvením na lidský jaderný protein, barvením na železo a detekcí lidské DNA v místě léze. Ukázali jsme, že buňky značené magnetickou protilátkou je možné sledovat magnetickou rezonancí. (cs)
