  • It was shown that treatment with FGF2 induced growth arrest in the G1 phase and partial de-differentiation of RCS chondrocytes. FGF2 also activated PLCg, protein kinase B, and Erk and p38 MAP kinases. Inhibition of FGFR3 and MEK1/2 antagonized FGF-2-mediated growth arrest. Expression of a dominant negative Ras resulted in a partial reversal of growth inhibition, while expression of constitutively activated Ras led to Erk-dependent growth arrest. At the molecular level, FGF2-induced growth arrest was followed by changes in cell cycle regulating complexes

  • Bylo prokázáno, že FGF2 indukuje zástavu růstu v G1 fázi buněčného cyklu a rediferenciaci chodrocytů RCS. FGF2 u těchto buněk také aktivuje PLCg, protein kinázu B, a Erk a p38 MAP kinázy. Inhibice FGFR3 a MEK1/2 u RCS buněk působí antagonicky na růstovou inhibici indukovanou FGF2. Ukázalo se, že exprese dominantně negativního Ras, částečně snižuje inhibici růstu vyvolanou FGF2. Zárověn, exprese konstitutivně aktivního Ras inhibuje růst těchto buněk v závislosti na aktivitě Erk. Současně s inhibicí růstu RCS buněk byly detekovány i změny v expresi a asociaci regulátorů buněnčného cyklu (cs)
