  • We tested 96 microsatellites and 10 single nucleotide polymorphisms for their allelic distribution in two subspecies of the house mouse, Mus musculus musculus and M. m. domesticus. Sixty-two microsatellites discriminated strain-specific differences among nine wild-derived %22musculus%22 and %22domesticus%22 and three %22classical%22 laboratory strains. For efficient genotyping, we optimized multiplex conditions using five microsatellites per polymerase chain reaction. All 10 single nucleotide polymorphisms were also optimized for simultaneous analysis in one reaction using SNaPshot multiplex. The uniform distribution of markers on autosomes and on the X chromosome makes these panels potentially useful tools for quantitative trait loci mapping of wild house mice.

  • Testovali jsme 96 mikrosatelitů a 10 SNP (single nucleotide polymorphism) markerů u dvou poddruhů myši domácí – Mus musculus musculus a M. m. domesticus. 62 z nich odlišuje kmenově specifické varianty markerů u devíti myších kmenů odvozených od divokých myší musculus a domesticus a tří klasických laboratorních kmenů. Pro maximálně efektivní genotypování jsme PCR zoptimalizovali tak, že v každé reakci běželo pět mikrosatelitů zároveň. Všech 10 SNP markerů bylo optimalizováno v jedné reakci pomocí SNaPshot multiplexu. Rovnoměrné rozmístění použitých molekulárních markerů na autosomech a X chromosomu dělá z tohoto panelu nástroj pro QTL mapování divokých myší. (cs)
