Description	V návaznosti na naši předcházející studii jsme se zaměřili na přiblížení mechanizmů apoptózy a změn buněčného cyklu v buňkách adenokarcinomu tlustého střeva HT-29 předinkubovaných s 5-LOX inhibitorem MK-886 a následnou fotodynamickou terapií s hypericinem. Intenzivní akumulaci buněk v G0/G1 fáze cyklu provázejí změny v hladinách cyklinu E a A, cdk-2 a pRb proteinu. Apoptóza je charakterizována výraznou aktivací kaspázy-3, -8 a -9. Analýza jednotlivých typů kyslíkových radikálů naznačuje, že předinkubace nádorových buněk s inhibitorem MK-886 ovlivňuje jejich distribuci. (cs) The concern of our present work, in accordance to our previous study, is a mechanism of apoptotic process and alterations in cell cycle of colon adenocarcinoma cells HT-29 pretreated with 5-LOX inhibitor MK-886 prior to photodynamic therapy with hypericin. Intensive accumulation of HT-29 cells in G0/G1 phase of cell cycle are accompanied with changes in cyklin E and A, cdk-2 and pRb protein levels. Significant activation of caspase-3, -8 and -9 are typical for intensive apoptotic process. Reactive oxygen species (ROS) analysis suggests an effect of MK-886 inhibitor over ROS types distribution.
