  • Using biochemical methods, the effect of two factors that might play a role in the mechanism of the biological activity of cisplatin at elevated temperatures (<37oC) have been examined. It has been shown that increased temperatures result in distinct alterations in the modification of the target DNA by cisplatin, and in the repair of these modifications. In vitro results support the view that the enhanced DNA-cross-linking efficiency of cisplatin and the lower efficiency of native DNA repair mechanisms at higher temperature play at least a partial role in the potentiation of the antitumor effects of cisplatin under conditions of mild hyperthermia.

  • Pomocí biochemických metod byl zkoumán vliv dvou faktorů, které mohou hrát roli v mechanismu biologické aktivity cisplatiny při zvýšených teplotách (<37oC). Bylo ukázáno, že zvýšené teploty mají za následek výrazné změny v modifikaci DNA cisplatinou a opravě těchto modifikací. Výsledky studií provedených in vitro podporují názor, podle kterého zvýšená účinnost vytvářet v DNA můstky a snížená účinnost mechanismů opravy DNA při zvýšených teplotách hraje přinejmenším částečnou roli v zesílení protinádorových efektů cisplatiny za podmínek mírné hypertermie.
