  • Kadmium patří mezi nejnebezpečnější látky znečišťující prostředí. Kadmiové ionty navozují apoptózu buněk tabáku (TBY-2). Fytochelatiny (PCs) jsou peptidy bohaté na cystein a jsou zahrnuty v detoxifikaci těžkých kovů v rostlinách a jsou syntetizovány při odpovědi na vystavení těžkým kovům. Využili jsme elektrochemickou analýzu (na rtuťové elektrodě v přítomnosti kobaltu) k monitorování akumulace PC v buňkách TBY-2. Po kultivaci TBY-2 v přítomnosti cytotoxických koncentrací kadmia, hladina PC vzrostla během pre-apoptické fáze a dosáhla limitující hodnoty v kultivačním čase shodným se spuštěním apoptózy. Hladina PC pozorována při sub-cytotoxických koncentrací kadmia (10 uM) byla asi třikrát nižší než byla pozorována pro 50 nebo 100 uM ionty kadmia po 5 dnech expozice. (cs)
  • Cadmium belongs to the most dangerous environmental pollutants. Cadmium ions caused tobacco cells (TBY-2) to enter apoptosis. Phytochelatins (PCs) are cysteine-rich peptides involved in detoxification of heavy metals in plants and are synthesized in response to the heavy metal exposure. We utilized electrochemical analysis (at mercury electrode in presence of cobalt) to monitor accumulation of PCs in TBY-2 cells. Upon TBY-2 cultivation in the presence of cytotoxic cadmium concentrations, the PC levels increased during the preapoptotic phase and reached a limiting value at cultivation times coinciding with apoptosis trigger. The PC level observed for a sub-cytotoxic cadmium concentrations (10 uM) was about three times lower then observed for the 50 or 100 uM cadmium ions after 5 days of exposure.
