| - Cílem práce bylo poskytnout nová data ke znalostem o mechanismech, jimiž rekombinantní lidský granulocytární kolonie stimulující faktor (rhG-CSF), rekombinantní lidský granulocytární-makrofágové kolonie stimulující faktor (rhGM-CSF) a myší rekombinantní granulocytární-makrofágové kolonie stimulující faktor (rmGM-CSF) zvyšují počty kolonií rostoucích z krvetvorných progenitorových buněk pro granulocyty a makrofágy (GM-CFC) v myší kostní dřeni. Bylo zjištěno, že jak rhG-CSF, tak rmGM-CSF stimulují proliferaci GM-CFC kombinací přímých efektů (přímé účinky na cílové buňky) a nepřímých efektů (účinky zprostředkované indukcí dalších cytokinů a růstových faktorů v organismu), zatímco v případě rhGM-CSF byly zjištěny pouze účinky nepřímé. - The aim of the work was to provide new data on the knowledge of mechanisms by which recombinant human granulocyte colony-stimulating factor (rhG-CSF), recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF), and recombinant mouse granulocyte-macrophage colony-stimulating factor (rmGM-CSF) enhance numbers of colonies growing from hematopoietic progenitor cells for granulocytes and macrophages (GM-CFC) in mouse bone marrow. It has been found that both rhG-CSF and rmGM-CSF stimulate proliferation of GM-CFC by a combination of direct effects (direct action on target cells) and indirect effects (action mediated by induction of other cytokines and growth factors in the organism) whereas in the case of rhGM-CSF only indirect effects have been observed.
