  • In the present study a series of thirty-two hydroxy and dihydroxy-PCBs (OH-PCBs) and PCB-derived quinones were prepared and evaluated for their in vitro potencies to downregulate gap junctional intercellular communication (GJIC) and to activate the aryl hydrocarbon receptor (AhR) and the estrogen receptor in liver and mammary cell models. The results show that oxygenated PCB metabolites are capable of multiple adverse effects, including gap junction inhibition, AhR-mediated activity and (anti)estrogenicity. The inhibition of GJIC by OH-PCBs represents a novel mode of action of both the lower-chlorinated and the persisting high-molecular-weight OH-PCBs.

  • V této práci byla studována série třiceti dvou hydroxy a dihydroxy-PCB (OH-PCB) a chinonů odvozených z PCB z hlediska jejich schopnosti potlačovat mezibuněčnou komunikaci typu “gap junction” (GJIC) a aktivovat receptor pro aromatické uhlovodíky (AhR) a estrogenní receptor v buněčných modelech jaterních a prsních buněk. Výsledky ukazují, že metabolity PCB jsou schopny způsobovat řadu škodlivých efektů, zahrnujíce v to inhibici GJIC, aktivitu zprostředkovanou AhR a (anti)estrogenitu. Inhibice GJIC pomocí OH-PCB představuje nový způsob účinků jak nízko-chlorovaných, tak perzistentních OH-PCB o vyšší molekulové váze. (cs)
