  • Glutathione in the reduced (GSH) and oxidized (GSSG) forms was studied by cyclic voltammetry. Tris(2-carboxyethyl)phosphine (TCEP) as the disulfide bond reductant and/or hydrogen peroxide as the sulfhydryl group oxidant were used. Cyclic voltammetry measurements, following the redox state of glutathione, were performed on a hanging mercury drop electrode (HMDE) in borate buffer (pH 9.2). It was shown that in aqueous solutions TCEP was able to reduce disulfide groups smoothly and quantitatively. The TCEP response at -0.25 V vs. Ag/AgCl/3 M KCl did not disturb the signals of the thiol/disulfide redox couple.

  • Glutathion v jeho redukované (GSH) a oxidované formě (GSSG) byl studován pomocí cyklické voltametrie. V experimentech byl využit TCPE pro redukci disulfidické vazby a peroxid vodíku pro oxidaci sulfhydrylové skupiny. CV měření byla provedena na HMDE v borátovém pufru, pH 9,2. Zjistili jsme, že ve vodném prostředí TCEP redukuje snadno a kvantitativně disulfidickou vazbu. Signál TCEP byl pozorován při potenciálu -0,25 V (proti Ag/AgCl/KCl elektrodě) a neovlivňoval signál redox páru thiol/disulfid. V práci je diskutován vznik signálů glutathionu GSH (při -0,44 a -0,37V) a GSSG (při -0,69 a -0,40 V). Stabilizace redukovaného stavu bílkovin a peptidů je velmi důležitá pro monitorování interakcí bílkovina-bílkovina, bílkovina-DNA a bílkovina-xenobiotika. (cs)
