  • Ačkoliv je řada polycyklických aromatických uhlovodíků (PAHs) považována za silné mutageny a karcinogeny, o jejich roli v nádorové promoci je známo relativně málo. Bylo prokázáno, že 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) může indukovat uvolnění krysích jaterních oválných epiteliálních buněk z kontaktní inhibice mechanismem pravděpodobně zahrnujícím aktivaci receptoru pro aromatické uhlovodíky (AhR). Řada PAHs jsou ligandy AhR, a také je známo, že působí jako přechodné induktory aktivity zprostředkované AhR. V této práci byly zkoumány efekty 19 vybraných PAHs na proliferaci konfluentní populace krysích jaterních epiteliálních buněk WB-F344. Výsledky této studie napovídají, že uvolnění z kontaktní inhibice může být součástí účinků PAHs aktivujících AhR, což vede k nádorové promoci; nicméně s tímto procesem by mohly také interferovat genotoxické účinky některých PAHs spojené s aktivací p53. It is known that 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) can induce release of rat hepatic oval epithelial cells from contact inhibition by a mechanism possibly involving the aryl hydrocarbon receptor (AhR) activation. Many PAHs are AhR ligands and are known to act as transient inducers of AhR-mediated activity. In this study, effects of 19 selected PAHs on proliferation of confluent rat liver epithelial WB-F344 cells were investigated. The results of the present study seem to suggest that a release from contact inhibition could be a part of tumor promoting effects of AhR-activating PAHs; however, the genotoxic effects of some PAHs associated with p53 activation might interfere with this process.
