Description
  • Chronic myeloid leukemia (CML) is caused by a BCR-ABL translocation in a primitive hematopoietic stem cell (HSC). There is evidence that the number of HSCs in the body is only loosely regulated, perhaps having a 'dead-band' of physiologically acceptable values. The existence of a dead-band suggests that low levels of HSC killing can be permanent, i.e. that low doses can cause permanent reductions in the total number of CML target cells and thus permanent reductions in the subsequent risk of spontaneous CML. Our preliminary calculations indicate that, due to dead-band HSC control, if HSCs are as low dose hypersensitive to killing as epithelial cells, reductions in spontaneous CML risk could cancel the low dose risks of induced CML, i.e. net lifetime CML risk could have a 'U' shaped low dose response.
