  • Neural stem cells (NSC) are promising source of cells for the treatment of central nervous system diseases. Access to regulatory signaling proteins present in low copies has been very restrictive until now due to scarcity of stem cell populations. Hence, we utilized the KinexTM Antibody Microarray analysis where profiles of the proliferating porcine NSC and differentiated counterparts were compared. Selected changes were verified by immunoblotting. Differentiated neural cells exhibited an increased level of RafB kinase, MAPKK 3, heme oxygenase 2 (HO2) and protein phosphatase 4C. In contrast, for the NSC were typical high level of G protein-coupled receptor-serine kinase 2 and enhanced phosphorylations of B-crystallin (S45), PKC (T655), PKC (S738/S742) together with eIF2 (S51). Immunocytochemistry confirmed expression profiles and intense cytoplasmic localization of HO2 in neurons but a weaker signal in glia. Phospho-S45 B-crystallin was localized in nuclei of differentiated cells.

  • Nervové kmenové buňky představují nadějný zdroj buněk pro léčbu onemocnění centrálního nervového systému. Výzkum regulačních signálních proteinů přítomných ve velmi malém počtu molekul byl dosud velmi limitován díky vzácnosti kmenových buněk. Proto jsme použili analýzu pomocí KinexTM protilátkových mikročipů k porovnání profilů nervových kmenových buněk a jejich diferencovaných protějšků a vybrané nalezené změny jsme potvrdili imunoblotem. Diferencované buňky vykazovaly zvýšené hladiny RafB kinasy, MAPKK 3, heme oxygenasy 2 (HO2) a protein fosfatasy 4C. Naopak pro kmenové buňky byla typická vysoká hladina G protein-coupled receptor-serine kinasy 2 a zvýšené fosforylace B-crystallinu (S45), PKC (T655), PKC (S738/S742) a eIF2 (S51). Podrobnější studie HO2 a fosfo-S45 B-crystallinu potvrdily výsledky mikročipů a intenzivní cytoplasmatickou lokalizaci HO2 v neuronech spolu se slabším signálem v gliích. Fosfo-S45 B-crystallin byl detekován v jádrech diferencovaných nervových buněk. (cs)
