  • Serin/threoninová kinasa Aurora A (AURKA) je jednou z klíčových molekul podílejících se na regulaci buňěčného cyklu u somatických buňěk. V této práci ukazujeme její úlohu v průběhu meiotického zrání myších oocytů jako modelu pro G2-M přechod. Studie založené na specifické inhibici molekul jako CDK1 (Roscovitine), PI3K ( LY-294002) a PKB (SH-6) indikují, že aktivace AURKy na MTOC je nezávislá na PI3K-.PKB a CDK1 signální dráze. Navíc úloha AURKy byla prokázána pomocí overexprese či deplece AURKA proteinu pomocí RNAi. Výsledky těchto experimentů jsou jasným důkazem toho, že AURKA se podílí nejen na znovuzahájení meiosy, ale i na pomnožení MTOC, tvorbě meiotického vřeténka či regulaci přechodu mezi metafází I a II.   • Aurora kinase A (AURKA), which is a centrosome-localized serine/threonine kinase crucial for cell cycle control in somatic cells. We report localization and functions of AURKA during meiotic maturation of mouse oocytes as a model system to study the G2 to M transition. Inhibitor studies using Roscovitine (CDK1 inhibitor), LY-294002 (PI3K inhibitor) and SH-6 (PKB inhibitor) reveal that activation of AURKA localized on MTOCs is independent on PI3K-PKB and CDK1 signaling pathways. Overexpression of AURKA leads to formation of an abnormal MI spindle,whereas RNAi-mediated reduction of AURKA interferes with resumption of meiosis and spindle assembly. Results of these experiments indicate that AURKA is a critical MTOC-associated component involved in resumption of meiosis, MTOC multiplication, proper spindle formation and the metaphase I-metaphase II transition.
