  • Partial and rarely complete spontaneous regression have been observed in cutaneous melanoma patients but little is known about the molecular mechanisms involved. The Melanoblastoma-bearing Libechov Minipig (MeLiM) is a suitable model to study the spontaneous regression because MeLiM pigs exhibit naturally occurring melanomas which regress completely 6 months after birth. In this study, we used suppression subtractive hybridization (SSH) to identify molecular determinants of melanoma regression within swine melanoma tissues and melanoma cell cultures. Several markers involved in cell-adhesion,-communication,-motility, signal transduction, negative regulation of cell proliferation and immune response were identified that correlated with melanoma regression. We identified the retinoic acid responder 1 gene (RARRES1) as a main actor of the regression in melanoma. For the most differentially expressed genes, we validated the results obtained by SSH with qRT-PCR and immunohistochemistry.

  • U pacientů s kožním melanomem byla pozorována částečná a velmi vzácně úplná spontánní regrese nádorů, ale o molekulárních mechanismech tohoto procesu je známo velmi málo. MeLiM (Melanoblastoma-bearing Libechov Minipig) model je vhodný pro studium spontánní regrese protože miniprasat MeLiM vykazují dědičně podmíněné melanomy, které kompletně regredují během 6 měsíců po narození. V této studii jsme použili pro detekci molekulárních determinant regrese melanomu v melanomové tkáni a v kultivovaných melanomových buňkách metodu supresní subtraktivní hybridizace (SSH). Bylo identifikováno několik markerů zapojených do buněčné adheze, komunikace a migrace, do signální transdukce a negativní regulace buněčné proliferace a do imunitní odpovědi, které korelovaly s regresí melanomu. Jako hlavní faktor byl poprvé u melanomu identifikován gen RARRES1 (retinoic acid responder 1). Pro nejodlišněji exprimované geny byly výsledky SSH ověřeny qRT-PCR a imunohistochemicky. (cs)
