  • Byla provedena analýza aktivace PKB (proteinkináza B) a CDK1 (cyklin-dependentní kináza 1) v průběhu zrání oocytů myši. Během zrání oocytů v podmínkách in vitro a in vivo fosforylace a aktivace PKB předcházela GVBD (rozpad zárodečného váčku). Tato aktivace byla dočasná a aktivita PKB výrazně poklesla po té, co téměř všechny oocyty prošly GVBD. Ošetření oocytů butyrolactonem I (inhibitor CDK1) prokázalo, že k aktivaci PKB dochází nezávisle na aktivitě CDK1. LY-294002, inhibitor fosfoinositid 3-kinázové-PKB signální dráhy, potlačil aktivaci PKB a CDK1 jakož i znovuzahájení meiózy. Během znovuzahájení meiózy PKB fosforylovaná na Ser473 byla lokalizována na jaderné membráně a na centrozómu, zatímco PKB fosforylovaná na Thr308 byla detekována pouze na centrozómu. Výskyt fosforylované PKB na centrozómu v průběhu GVBD naznačuje, že PKB má významnou funkci při počáteční aktivaci CDK1. (cs)
  • Activation of PKB (protein kinase B) and CDK1 (cyclin-dependent kinase 1) during maturation of mouse oocytes was analysed. PKB phosphorylation and activation preceded GVBD (germinal vesicle breakdown) in oocytes maturing either in vitro or in vivo. Activation was transient and PKB activity was markedly reduced when virtually all of the oocytes had undergone GVBD. Treatment of oocytes with butyrolactone I, a specific inhibitor of CDK1, revealed that PKB activation is independent of CDK1 activity. LY-294002, an inhibitor of phosphoinositide 3-kinase–PKB signalling, suppressed activation of PKB and CDK1 as well as resumption of meiosis. During resumption of meiosis, PKB phosphorylated on Ser473 is associated with nuclear membrane and centrosome, whereas PKB phosphorylated on Thr308 is localized on centrosome only. The presence of phosphorylated PKB on centrosome at the time of GVBD suggests its important role for an initial CDK1 activation.
