Description
  • Fas (CD95/APO-1) belongs to the TNF receptor (TNRF) family. Fas ligand binding followed by Fas-receptor oligomerisation leads to formation of a deathincluding signal cpmlex starting with recruitment of the Fas-adapter protein (FADD). Components of this initiation complex were correlated with apoptotic cells, detected by specific DNA fragmentation and morphological criteria. Apoptotic cells can be detected throughout the embryonic development of molar teeth. Restricted temporospatial distribution suggests several important roles for apoptosis in tooth morphogenesis. However, the mechanisms employed in dental apoptosis remain unclear. According to the immunohistochemical data, Fas-mediated signalling may have a triggering or enhancing role in dental apoptosis. This remains to be functionally confirmed.

Po navázání ligandu na receptor dojde k jeho oligomerizaci zapojení Fas-adaptorového proteinu (FADD) a formaci apoptózu indukujícího signalizačního komplexu. Komponenty tohoto iniciačního komplexu (Fas, Fas-L, FADD) byly srovnány s apoptotickými buňkami, určenými na základě fragmentace DNA a specifických morfologických kritérií. Apoptózu lze detekovat během celého vývoje stoliček. časoprostorové rozložení apoptotických buněk naznačuje, že apoptóza má důležitou roli v morfogenezi zubních základů, i když mechanismy zapojené do tohoto procesu zůstávají nejasné. K popsání lokalizace Fas, Fas-L a FADD molekul a apoptózy během vývoje první stoličky a jejich vzájemnému srovnání byla použita imunohistochemie na frontálních řezech hlavami hraboše mokřadního ve stádiu embryonálního vývoje 13,5-15,5 dne.
