Description
| - Olej z černého rybízu je bohatý na vícenenasycené mastné kyseliny. Cílem studie bylo získat částečnou hydrolýzou oleje směs volných mastných kyselin (VMK) odlišného složení, než měl výchozí olej. Roztok oleje a vody v superkritickém CO2 protékal reaktorem s pevným ložem Lipozymu, 1,3-specifické lipasy imobilizované na makroporézní pryskyřici za podmínek: teplota 40 C, tlak 15-28 MPa, postupná rychlost 0,1-0,7 mm/s. Složení hydrolyzátu bylo analyzováno kolorimetrickou metodou, PTLC, GC, HPLC a LC-NMR. Maximální reakční rychlost 2.6 mmol/s (kg enzymu) byla dosažena při maximální ryclosti toku a maximálním tlaku. Frakce VMK obsahovala víc nasycených kyselin než olej, ve shodě s faktem, že nasycené kyseliny jsou v triacylglycerolech rostlinných olejů v pozicích sn-1 a sn-3. Koncentrace gama-linolenové kyseliny v FFA byla o řád nižší než v oleji. (cs)
- Blackcurrant seed oil is rich in polyunsaturated acids. The aim of the study was to obtain by partial hydrolysis a mixture of free fatty acids (FFA) of different composition than that of the hydrolyzed oil. Solution of oil and water in scCO2 flowed through a packed bed reactor filled with Lipozyme, a 1,3-specific lipase immobilised on a macroporous ion-exchange resin. The temperature was 40 C, pressure 15-28 MPa, and superficial velocity 0.1-0.7 mm/s. The hydrolysate composition was determined using colorimetric method, PTLC, GC, HPLC, and LC-NMR. The maximum reaction rate 2.6 mmole/s (kg enzyme) was achieved at maximum flow velocity and maximum pressure. The FFA fraction contained higher percentage of saturated fatty acids than the hydrolyzed oil, in accordance with the fact that saturated acids occupy sn-1 and sn-3 positions in triacylglycerols of vegetable oils. Concentration of gamma-linolenic acid in FFA fraction was by order of magnitude lower than its concentration in the oil.
