  • We demonstrate that the chemical modification of TRPV1 receptor by both reducing and oxidizing agents leads to an increased response to heat in HEK293T cells (human embryonic kidney) transfected by gene of TRPV1 receptor. Using site-directed mutagenesis we showed that the cysteine at position 621 located extracellularly in the pore region contributes to the DTT induced potentiation of heat-activated currents mediated by TRPV1, and we found that extracellular cysteines (C616, C621, C634) are not involved in TRPV1 modulation by oxidizing agents. Based on the chromatographic sepraration we demonstrate that oxidizing substances can influence the concentration of TRPV1 agonists and thus indirectly activation of receptor

  • Práce se zabývá vlivem redukčních a oxidačních látek na aktivaci vaniloidního receptoru a ukazuje, že jak redukční, tak oxidační látky zvyšují membránové proudy vyvolané zvýšením teploty aplikovaného roztoku na HEK293T buňky (lidské embryonální ledvinné) transfekované genem TRPV1 receptoru. Pomocí cílené metageneze byl určen cystein 621, lokalizovaný extracelulárně v oblasti póru iontového kanálu, který se významně podílí na potenciaci odpovědí vyvolaných teplem působením redukční látky dithiothreitol, a bylo prokázáno, že extracelulární cysteiny (C616, C621, C634) se neúčastní senzitizace odpovědí na teplo vlivem oxidačních látek. Pomocí vysokoúčinné kapalinové chromatografie bylo prokázáno, že oxidační látky ovlivňují koncentraci agonistů TRPV1 receptoru a tím nepřímo jeho aktivaci (cs)
