  • The aim of this study was to ascertain whether GPDH activity could be used as an index of thyroid status during chronic experiments over several months. We therefore analyzed GPDH activity in liver mitochondria of female inbred Lewis rats with thyroid status altered for 2 to 12 months. Hyperthyroid state was maintained by triiodothyronine (T3) or thyroxine (T4) administration, while methimazole was employed for inducing hypothyroidism. We found a seven- and three-fold increase of GPDH activity in female rats after T3 and T4 administration, respectively, compader to euthyroid females, whereas administration of methimazole reduced the enzyme activity almost to one-third of the euthyroid values. We conclude that the level of the rat liver GPDH activity could serve as a useful marker for evaluation of hyperthyroid and hypothyroid status in chronic long-lasting experiments on female inbred Lewis rats

  • Cílem práce bylo zjistit, zdali GPDH aktivita může být použita jako měřítko tyroidního stavu v průběhu chronických experimentů trvajících několik měsíců. Analyzovali jsme proto GPDH aktivitu v jaterních mitochondriích samic inbredního kmene Lewis potkana se změněným tyroidním stavem v trvání 2-12 měsíců. Hypertyroidní stav byl udržován injekcí trijodtyroninu (T3) nebo tyroxinu (T4), zatímco methimazol byl použit na vyvolání hypotyroidismu. Zjistili jsme 7- a 3násobný vzestup GPDH aktivity po aplikaci T3 resp. T4 ve srovnání s eutyroidními zvířaty, zatímco methimazol redukoval enzymovou aktivitu téměř o dvě třetiny ve srovnání s hodnotami eutyroidních zvířat. Na základě těchto výsledků můžeme uzavřít, že stanovení jaterní GPDH aktivity může být používáno jako užitečný marker pro hodnocení hypertyroidního nebo hypotyroidního stavu v chronických dlouhotrvajících experimentech (cs)
