  • Whole-cell patch-clamp recording from human embryonic kidney 293 cells expessing NR1-1a/NR2B receptors revealed that neurosteroid pregnenolone sulfate (PS) (300mm), when applied to resting NMDA receptors, potentiates the amplitude of subsequent responses to 1 mM glutamate fivefold and slows their deactivation twofold. The same concetration of PS, when applied during NMDA receptor activation by 1nM glutamate, has only a small effect. These results suggest that the PS is a modulator of NMDA receptor Po, the effectiveness of which is lowered by glutamate binding

  • Rekombinantní NR1-1a/NR2B receptory byly exprimovány v lidských embryonálních ledvinných (HEK293) buňkách, ze kterých byly snímány odpovědi pomocí metody patch-clamp. Zjistili jsme, že pokud preaplikujeme pregnenolon sulfát (PS; 300 mM) na agonisty neaktivované NR1-1a/NR2B receptory, dochází k pětinásobné potenciaci odpovědí těchto receptorů a zároveň ke dvojnásobnému zpomalení doby deaktivace. Stejná koncentrace PS aplikovaná na NR1-1a/NR2B receptory aktivované předchozí aplikací glutamátu potencuje odpovědi těchto receptorů minimálně. Naše výsledky ukazují, že PS je modulátor pravděpodobnosti otevření NR1-1a/NR2B receptorů, jehož účinnost je snížena vazbou glutamátu (cs)
