  • The aim of this study was to analyze the occurrence of extended-spectrum beta-lactamase (ESBL)- and plasmid-mediated quinolone resistance (PMQR)-positive Escherichia coli isolated from cormorants (Phalacrocorax carbo) and mallards (Anas platyrhynchos) in the central Europe, and to determine the importance of these birds in spreading of these bacteria. The samples were cultivated parallelly on MacConkey agar (MCA) with cefotaxime (2 mg/L) to isolate ESBL-producing E. coli and MCA with ciprofloxacin (0.05 mg/L) to isolate fluoroquinolone-resistant E. coli. PCR was used to detect specific antibiotic resistance genes. Plasmids carrying ESBL and/or PMQR genes were transferred by conjugation or transformation and characterized by PCR-based replicon typing and restriction analysis. From waterbirds, ten ESBL-producing E. coli with the blaCTX-M-15 or blaCTX-M-27 genes were isolated. In most isolates with blaCTX-M-15, the aac(6')Ib-cr gene was also detected. The bla genes were harbored by F and I1 conjugative plasmids. PMQR genes were detected in 25 E. coli. Six strains carried aac(6')Ib-cr and nineteen strains were positive for qnrS1. The genes were located on transferable plasmids of incompatibility groups N or X2. The finding of CTX-M-type ESBLs and PMQR genes in these animal species indicates rapid spread of bacteria with these resistance genes in the environment.
