  • Hemorrhagic enteritis virus (HEV) belongs into Siadenovirus genus , family Adenoviridae and infects avian species. HEV causes a widespread enteral disease of turkey characterized by depression, hemorrhagic diarrhea, splenomegaly and immunosuppression in turkeys from four weeks of age. The target tissue for virus replication is lymphatic system. The virus replicates in B cells and macrophages and is concentrated in large amount in the spleen. The aim of this work was to express recombinant hexon protein of hemorrhagic enteritis virus, to establish the diagnostic value of this protein for serological detection of antibodies in turkey serum samples and to assess seroprevalence of the disease in the Czech Republic. The gene fragment coding N' terminal part of hexon protein was PCR amplified and expressed in bacterial expression system. Serological reactivity of recombinant hexon protein was assessed on a panel of 98 turkey sera by immunoblotting. Serum samples originated from turkey farms in the Czech Republic with the history of hemorrhagic enteritis virus infection, from farms with animals free of infection and from turkey farms after HEV vaccination. The results with HEV hexon protein obtained by immunoblotting were compared with ELISA kit (Synbiotics). Both immunoblot and commercial ELISA kit gave similar results with tested sera. Immunoblot using recombinant hexon protein proved useful for detecting turkey flocks infected with hemorrhagic enteritis virus.
