Description
  • The growth curve is one of the most important characteristics of batch cultivation procedure in plant biotechnology. There are several ways how to assess this parameter. Cells can be counted directly using a counting chamber, but this is time-consuming work and in some cases almost impossible owing to cluster formation. Indirect methods like optical density, sedimentation characteristics etc. are not suitable for plant cell suspensions due to morphological changes during cultivation period. Recent papers suggest that intracellular esterase activity of plant cultures depends only on the amount of living cells. Thus determination of esterase activity seems to be a good indirect method to obtain cell density (living cells only) and finally growth curve if calibrated using direct counting and conventional viability test. The main goal of our work was to develop an inexpensive assay for esterase activity.
