Description
| - Embryogenic cell suspension cultures from hypocotyl segments of cucumber (Cucumis sativus L.) have been initiated by 7 days exposure to Murashige and Skoog medium supplemented with 3% sucrose, 2.2 mg l-1 2,4-dichlorophenoxyacetic acid, and 100 mg l-1 casein hydrolyzate. The suspension cultures were maintained in the same medium with 1.1 mg l-1 2,4- dichlorophenoxyacetic acid only on a horizontal shaker at 28°C, and 16/8 h photoperiod, subcultivated every week. The cell suspension cultures contained heterogeneous arrays of several different types of single cells (size < 200 µm) and cell clusters (size ? 200 µm). The average generation time was 3 days. The somatic embryos developed after transfer from auxin-containing medium to auxin-free medium. Embryogenic capacity of the cell lines varied between 35 and 470 somatic embryos per 1 ml of cell suspension.
