  • Cílem této práce bylo detekovat stříbrné ionty pomocí různých elektrochemických nástrojů. Uhlíkové elektrody byly testovány jako pracovní elektrody. Optimalizované postupy byly využity pro stanovení stříbra (I) na tabáku BY-2 buněk a ryby guppy, ketrá byla zvystavená stříbrnými ionty. Dále jsme optimalizovali základní experimentální parametry pro vysoce účinnou kapalinovou chromatografii s elektrochemickou detekcí (HPLC-ED) pro stanovení stříbrných iontů. Nejvhodnějšími parametry pro stanovení stříbrných iontů byly tyto - mobilní fáze 0,2 M octan pufr (pH 4,0) a její průtok: 0,5 ml / min, potenciál ochranné cely: 0 mV,potenciál pracovní elektrody: 200 mV, aktuální R: 1 UA. Detekční limit byl 20 nmol/dm3. Obsah stříbrných iontů zjištěných u pokusných BY-2 buněk se zvyšoval s koncentrací stříbra a dobou působení. (cs)
  • The aim of this work was to detect silver ions by using various electrochemical instruments. Carbon electrodes were tested as working electrodes. The optimized procedures were utilized for determination of silver(I) at BY-2 tobacco cells and guppy fishes treated with the silver(I) ions. Moreover we optimized the basic experimental parameters of the high performance liquid chromatography with electrochemical detection (HPLC-ED) to determine silver(I) ions. The most suitable parameters for the determination of silver ions were as follows--mobile phase 0.2M acetate buffer (pH 4.0) and its flow rate: 0.5 ml/min, guard cell potential: 0 mV, working electrode potential: 200 mV, current R: 1 uA, time filter: 2 s. The detection limit obtained was 20 nmol/dm3. The content of silver(I) ions determined in the treated BY-2 cells enhanced with increasing dose of the silver(I) ions and time of the treatment.
