Description
| - Několik českých kultivarů hrachu bylo transformováno geny ovlivňujícími odolnost vůči škůdcům a patogenům. Tři konstrukty obsahující gmSPI-2 (Galleria mellonella silk proteinase inhibitor) byly připraveny pro transformaci hrachu pomocí Agrobakteria. (cs)
- A set of Czech pea cultivars was genetically modified with genes effecting resistance to pests and pathogens. Three constructs containing gmSPI-2 gene (Galleria mellonella silk proteinase inhibitor) were prepared for Agrobacterium-mediated transformation. Three types of regeneration system of pea derived from mature seeds (induction of trimmed seeds -- TS, cotyledonary nodes -- CN), and immature embryos (IE) were used for application of transformation protocols based on work of Švábová et al. (2005, 2007). After cocultivation, selection and regeneration steps, the potentially transformed plants were cultivated ex vitro, acclimatized and planted in a culture room to yield seed generation T1. Samples of leaf tissues were taken to test gus and bar gene activities histochemically, and then by molecular methods (PCR).
