| - The main aim of our work was to utilize adsorptive transfer stripping technique coupled with differential pulse voltammetry Brdicka reaction to determine thiols in early somatic embryos of Spruces (ESEs) treated with 0, 250 and 500 uM concentrations of cadmium(II) ions as EDTA chelate for twelve days. If we analysed standard of GSH by Brdicka reaction, we obtained four signals called as RS2Co, Cat1, Cat2 and Cat3. For the qunatification purposes we used Cat3 signal. The linear part of this dependence was observed in the concentration range of 0 to 10 umol/L of GSH (y = 0.6419x + 0.1628; R2 = 0.9961). After that, we analysed the ESEs sample by AdTS DPV Brdicka reaction. Potential of RS2Co signal of ESEs treated by Cd-EDTA was shifted to more positive potential (about 200 mV) in comparison with potential of RS2Co signal of non-treated ESEs. It clearly follows from the results obtained that content of GSH was proportional to dose of Cd-EDTA and time of treatment.

- Cílem této práce bylo využít adsorptivní přenosovou techniku kombinovanou s diferenční pulsní voltametrií a Brdičkovou reakcí ke stanovení thiolů v časných somatických embryích smrku vystaveným působení 0, 250 a 500 uM koncentarce kademnatých iontů v podobě EDTA chelátu po dobu 12 dnů. Když byl analyzován standart GSH pomocí Brdičkovy reakce, byly získány 4 signály nazvané jako RS2Co, Cat 1, Cat 2, Cat 3. Pro úmysl látky kvantifikovat byl vybrán signál Cat 3. Lineární část této závislosti byla v rozmezí 0-10 umol/l GSH. (y = 0.6419x + 0.1628, R2 = 0.9961). Poté byly embrya analyzovány. Potenciál signálu RS2Co se posunul k pozitivnějším potenciálům. (o 200 mV) v porovnání s potenciálem signálu neovlivněných embryí. Obsah GSH byl úměrný k dávce Cd a době expozice. (cs)
