Description
| - Jako modelová rostlina v této práci byl vybrán tabák (Nicotiana tabacum) transformovaný pomocí Agrobacterium tumefaciens. Konstrukt byl schopný exprimovat kvasinkový metalothionein.. Předmětem zájmu bylo stanovení množství exprimovanéo metalothioneinu v rostlinných tkáních. Pro tento účel jsem použily optimalizovanou techniku nazvanou adsorpční přenosová technika spojená s diferenční pulsní voltametrií- Brdičkova reakce. Zjistil jsme, že rostlin syntetizovali rozdílná množství metalothioneinu. (cs)
- The model plant in this work tobacco (Nicotiana tabacum) was chosen to be transformed by Agrobacterium tumefaciens (Macek et al., 2002). The construct were then able to express yeast metallothionein gene. Nevertheless we were interested in the issue, if we would be able to quantify the expressed MTs. For these purposes we suggested and optimized electrochemical method called adsorptive transfer stripping technique coupled with differential pulse voltammetry -- Brdicka reaction (Petrlova et al., 2006). We found out that plant were able to synthesized from tens to hundreds ng MT per g of fresh weight.
