| - V této práci byly charakterizovány morfologické vlastnosti makrofágů v průběhu iniciace a rezoluce zánětlivé odpovědi mléčné žlázy skotu. Studie byla provedena na 20 mléčných žlázách 5 virginních jalovic s použitím světelné mikroskopie nativních a barvených buněk a pomocí transmisní elektronové mikroskopie (TEM). Zánětlivá reakce mléčné žlázy byla indukována intramammární instilaci PBS. Bylo zjištěno, že obě fáze zánětlivé reakce -- iniciální fáze a fáze rezoluce - jsou charakteristické přítomností různých morfologicky diferentních forem makrofágů. Během iniciální fáze zánětlivé odpovědi tvořily majoritní podíl populace makrofágů monocytům podobné makrofágy, jež představovaly čerstvě migrované buňky. Monocytům podobné makrofágy byly 12 -- 15 um veliké, sférického až ovoidního tvaru s homogenní, jemně zrnitou cytoplasmou s bohatým GK, četnými mitochondriemi a bez lysosomů. Jejich jádro mělo ledvinovitý tvar s tmavým chromatinem na periferii. Již během iniciální fáze byly zaznamenány makrofágy s fagocyt (cs)
- This work characterises macrophage morphological features during initiation and resolution of the inflammatory response by the bovine mammary gland. The study has been carried out in 20 mammary glands of 5 virgin heifers by using light microscopy of natural and stained cells and by using transmissive electron microscopy (TEM). The inflammatory reaction was induced by an intramammary administration of PBS. It has been found that both the initial as well as the resolution phases of the inflammatory reaction are characteristic of the presence of various morphologically different macrophage forms. During the initial phase of the inflammatory response, the major proportion of the macrophage population consisted of monocyte like macrophages, which represented newly migrated cells. These macrophages were 12 -- 15 um in size, with spherical or ovoidal shapes, and contained homogenous, fine-granular cytoplasm rich in Golgi complexes, numerous mitochondria, and no lysosomes.
