  • Smrk ztepilý (Picea abies (L.) Karst.) je pro horské lesy Evropy mimořádně důležitou dřevinou nejen z lesnického a dřevařského hlediska, ale také pro svou krajinotvornou a ekologickou funkci. Proliferující embryogenní kulturu je možné rychle množit, udržovat dlouhou dobu, nebo ji specifickým způsobem zmrazit a uchovat pro pozdější použití. K odlišení, charakterizaci a testování jedinců a jejich potomků významnou měrou přispívá rychle se rozvíjející oblast metod založených na molekulárních markerech. Jednotlivé klony dlouhodobě kultivovaných somatických embryí dobře rozlišujeme pomocí RAPD markers, které pro zjednodušení práce a lepší reprodukovatelnost výsledků konvertujeme na klonově-specifické SCAR markery. Tyto výsledky se snažíme potvrdit pomocí metod AFLP and analýzy mikrosatelitů. (cs)

Karst.) is an exceptionally important European mountain woody species, not only in terms of forestry and wood technology, but also because of its landscape-forming and ecological functions. The proliferating embryogenic culture can be rapidly propagated, maintained for a long time, or frozen in a specific way and kept for further use. Rapidly developing methods based on molecular markers considerably contribute to the differentiation, characterization and testing of individuals and their progeny.Individual clones of long-term-cultivated spruce somatic embryos were easily distinguished by RAPD markers. For better reproducibility RAPD results were converted into clone-specific SCAR markers. These results are now tested using the most powerful methods of AFLP and microsatellite analysis
