  • Mnozí členové nadrodiny cytochromu P450 (CYP) jsou inducibilní po expozici různým xenobiotikům v lidských játrech. Dosáhli jsme značných mechanistických poznatků těchto dějů v hepatocytech a několik ligandy-aktivovaných transkripčních faktorů bylo identifikováno v uplynulých dvou desetiletích. Rodiny CYP1, CYP2 a CYP3 zapojené do metabolismu xenobiotik jsou rovněž exprimovány v extrahepatálních tkáních (např. střevě, mozku, ledvinách, placentě, plících, nadledvinkách, slinivce, kůži, mléčné žláze, děloze, vaječnících, varlatech a prostatě). Jelikož exprese většiny Isidorem se jeví býti velmi nízká v extrahepatálních tkáních ve srovnání s expresí v játrech dospělého člověka, úloha těchto enzymů v úhrnném metabolismu a celkové clearance je minoritní. Nicméně, bazální exprese a up-regulace extrahepatálních CYP enzymů může významně ovlivnit místní distribuci xenobiotika nebo endogenní sloučeniny v periferních tkáních a takto modifikovat jejich farmakologické/toxikologické účinky nebo ovlivnit absorpci xen (cs)

We have gained considerable mechanistic insights into these processes in hepatocytes and multiple ligand-activated transcription factors have been identified over the past two decades.
