Description
  • Metoda embryo-rescue byla využita při interspecifické hybridizaci Cucumis anguria a C. zeyheri. Embrya byla extirpována z plodů 1 - 6 týdnů po opylení. C. anguria byla použita jako mateřská komponenta, C. zeyheri jako otcovská. Pro kultivaci izolovaných embryí byla použita různá média. Média s kokosovým mlékem byla nejvhodnější pro klíčení nezralých embryí, média s kyselinou askorbovou a caseinhydrolyzátem byla vhodná pro následnou organogenezi. Byly získany rostliny, jejichž hybridní charakter byl potvrzen jak morfologicky (pomocí vybraných znaků), tak biochemicky (pomocí isozymové analýzy). Byly určeny 3 typy hybridů (H1.1., H1.2., H2.).   • Embryo-rescue was used to facilitate interspecific hybridization of Cucumis anguria L. and C. zeyheri Sond. (Cucurbitaceae). Embryos were excised from developing fruits in one week intervals for six weeks after hand pollination. Various culture media were compared. Media containing coconut water were the most suitable for initial germination, and a medium with ascorbic acid provided for the best embryo development and plant recovery. Viable plants were obtained from embryos, and their hybrid character was observed by visual inspection of several morphological traits and then confirmed by isozyme analyses. These plants showed characteristics from both parents, and analysis of the leucine aminopeptidase (LAP) locus revealed three hybrid types (i.e. H1.1, H1.2 and H2).
